Rapid N-Glycan Analysis Sample Prep on an Automatable Micro Chromatography Platform

Glycosylation can critically affect efficacy and pharmacokinetics of biotherapeutics, including antibodies. N-glycan profile analysis is currently performed by CE, HPLC or LC/MS, but sample preparation is a major bottleneck. We have developed a new N-glycan preparation procedure utilizing a micro-chromatography platform compatible with standard automation. The procedure includes glycoprotein purification from crude samples, rapid enzymatic N-glycan release, instant fluorescent labeling and glycan cleanup, and can process up to 192 samples in less than 3 hours.
